How to Reconstitute Peptides with Bacteriostatic Water
Reconstitution is the single most error-prone step in a peptide research protocol. This guide walks through the exact procedure using bacteriostatic water, plus the concentration math researchers get wrong most often.
What you need
- Lyophilised peptide vial, stored refrigerated.
- Bacteriostatic water (0.9 percent benzyl alcohol preserved).
- Sterile insulin or 1ml syringe with fine-gauge needle.
- Alcohol swabs.
Do not use plain sterile water or tap water. Only bacteriostatic water preserves the reconstituted peptide for multi-week use.
Step-by-step
- Bring the vial to room temperature for 10 minutes. Cold vials can crack.
- Swab both rubber stoppers with alcohol and let dry.
- Draw the chosen volume of bacteriostatic water (see math below).
- Inject the water slowly against the vial wall, not directly onto the powder.
- Do not shake. Gently swirl until the powder fully dissolves. This can take a minute.
- Label the vial with reconstitution date and concentration.
- Refrigerate immediately.
The concentration math
Peptide amount in mg divided by water volume in ml equals concentration in mg per ml. For a 10mg BPC-157 vial reconstituted with 2ml water, you get 5 mg/ml. On a standard U-100 insulin syringe, each unit mark equals 0.01ml, so 10 units contains 0.5mg peptide at that concentration.

Storage after reconstitution
- Refrigerate at 2 to 8 degrees C.
- Use within 4 weeks for most peptides.
- Do not freeze reconstituted solution, freezing damages tertiary structure.
- Protect from direct light.
Common mistakes
- Shaking the vial, causes foaming and can denature the peptide.
- Injecting water directly onto the powder cake at high speed.
- Reconstituting with normal sterile water, then storing beyond a day.
- Forgetting to label concentration, guaranteed dosing error later.
